阿魏酸川芎嗪后处理对大鼠心肌缺血再灌注损伤的影响

         

摘要

目的 观察阿魏酸川芎嗪(LF)后处理对大鼠心肌缺血再灌注损伤的影响,并探讨其可能机制.方法 48只雄性SD大鼠随机分成假手术组(SM组)、缺血再灌注组(I/R组)、川芎嗪组(LZ组)和LF组.采用结扎左冠状动脉前降支30 min再灌注120 min的方法复制心肌缺血再灌注模型.SM组只穿线不结扎,LZ、LF组分别于再灌注前颈静脉注射LZ和LF 40mg/kg.记录血流动力学参数,测定血清超氧化物歧化酶(SOD)、丙二醛(MDA),测定心肌梗死面积、心肌细胞凋亡指数、Fas蛋白表达等.结果 LZ和LF能明显加快心率、升高左室收缩末压和±dp/dtmax、降低左室舒张末压,提高血清SOD活性、减少MDA含量,缩小心肌梗死面积,降低心肌细胞凋亡指数,减少Fas蛋白表达,与I/R组比较差异有统计学意义(P<0.01);上述指标LF作用强于LZ,差异有统计学意义(P<0.05).结论 LF后处理可减轻在体大鼠心肌缺血再灌注损伤,其机制与清除氧自由基、抑制脂质过氧化反应和抑制细胞凋亡有关.%Objective To investigate the effects of ligustrazine ferulate (LF) postconditioning on myocardial ischemia reperfusion injury in rats,and explore the related mechanisms. Methods Forty-eight male rats were randomly divided into sham-operation (SM) group,ischemia reperfusion (I/R) group,ligustrazine (LZ) group,LF group. The rat model of myocardial I/R was established by occlusion of the left anterior descending coronary artery for 30 minutes and reperfusion for 120 minutes. Rats in LZ group and LF group received jugular vein injection of LZ and LF (40 mg/kg) before reperfusion. The rats of SM group were given only loose suture around IAD. The parameters of haemodynam-ics was record; superoxide dismutase (SOD), malondialdehyde (MDA ) in serum, myocardial infarct size, myocardial apoptosis index and the expression of Fas protein were measured. Results Compared with I/R group,HR was accelerated,LVESP and 眃p/dtmax was enhanced, LVEDP was reduced,SOD activity was increased,MDA content was decreased in LF group and LZ group (P < 0.01 );in addition,myocar-dial infarct size, myocardial apoptosis index and the expression of Fas protein all were reduced (P < 0.01 ). The effect of LF is stronger than LZ about aforesaid indicators, the difference had statistical significance (P < 0.05). Conclusion LF postconditioning can protect against myocardial I/R injury,and the protective effect may be related to the scavenging of oxide free radical,which further restrain the reaction of lipid peroxidation and apoptosis.
