苎麻叶总绿原酸提取工艺研究

             

摘要

[Objective] To extract chlorogenic acid from ramie leaves by ultrasonic method, and set different conditions to detect the extraction yield so as to obtain the optimum extraction condtions. [ Method ] The first quarter ramie leaves were firstly dried, and then extracted by ultrasonic method with ethanol concentration, material - liquid ratio, temperature, pH and extracting time as the single factor ,according to the single factor experimental result, three factors of ethanol concentration, material-liquid ratio and pH were chosen to design the orthogonal test. [ Results ] The extraction effect was greatly impacted by each single factor, and it was the best under the room temperature and 2 h extraction time; the orthogonal results showed that, the optimum extraction conditions were 40% ethanol, pH 5 and 1: 16 material-liquid ratio. [Conclusion] Ramie leaves contained 0.745% of chlorogenic acid. It is convenient to adopt ultrasonic method with a fine extraction effect.%[目的]采用超声波法提取苎麻叶片绿原酸,设定不同提取条件测定含量并得出最优提取条件.[方法]对苎麻头麻叶片烘干处理后,以一定浓度乙醇为提取液,设定乙醇浓度、料液比、温度、pH、提取时间为单因素,采用超声波法提取,并根据单因素实验结果,选取乙醇浓度、料液比、pH三个因素设计正交实验.[结果]各单因素对提取效果影响较大,温度设定常温,提取时间2h最佳;正交结果表明,最优提取条件为40%乙醇、pH =5、料液比1∶16.[结论]苎麻叶中含有0.745%的绿原酸,采用超声波法提取效果好,方法简便易行.
