苯酚-硫酸法测定慈姑中多糖的含量

摘要

目的 建立测定慈姑中多糖含量的方法.方法 采用水提醇沉法提取慈姑多糖,并用苯酚-硫酸法对所提慈姑多糖含量进行测定,测定波长490 nm.结果在范围20~100μg内呈良好线性关系,r2=0.9928,平均回收率为104%,RSD=4.72(n=5),且供试溶液在2 h内显色稳定.结论 苯酚-硫酸法测定慈姑中多糖含量,方法灵敏、重现性好,适用于慈姑多糖含量测定.%Objective To establish a method to measure the content of sagittaria polysaccharide. Methods Water extract - alcohol precipitation method was to extract sagittaria polysaccharide,and test the content of polysaccharides from sagittaria with the method of H2SO4 - Phenol,wavelength was read at 490 nm. Results There was a good linear relationship between 20~100 μg, r2=0.9928, average recovery was 104%, and RSD was 4.72(n = 5), the colour of the test solution is stable within 2 h. Conclusion H2SO4- Phenol method is sensitive, reproducible and suitable for the determination of the polysaccharide content of sagittaria.
