HPLC法测定一匹绸中东莨菪苷和东莨菪内酯的含量

摘要

A HPLC method was established for determination contents of Scopolin and Scopoletin in Argyreia acuta Lour.The separation was performed on ODS-C18 column,with gradient elution in methanol-0.2％ H3PO4 solution at the flow rate of 0.8 mL/min.The column temperature was 30 ℃,and the detection wavelength was 346 nm.The results showed that Scopolin and Scopoletin had good linear relationships in range of 0.019 2～0.096 0 mg/mL (r=0.999 9) and 0.004 48～0.022 40 mg/mL (r=0.999 5).The methodological results were in line with requirements.The method is simple and accurate,with good precision and repeatability,which could be used for the quality control of Argyreia acuta Lour.%建立了一匹绸中东莨菪苷和东莨菪内酯的含量测定方法.色谱柱为ODS-C 18,用甲醇-0.2％磷酸梯度洗脱,流速为0.8 mL/min,柱温30℃,检测波长为346 nm.东莨菪苷、东莨菪内酯0.019 2～0.096 0mg/mL (r=0.999 9)、0.004 48～0.022 40 mg/mL(r=0.999 5)呈现良好的线性关系,方法学验证结果符合要求.本方法简便准确,精密度和重复性好,可作为一匹绸药材的含量检测方法.
