上皮性卵巢癌组织中 ATG5、Beclin1与P53的表达及相关性研究

         

摘要

目的:检测自噬相关基因 ATG5、Beclin1与凋亡相关基因 P53在上皮性卵巢癌、良性卵巢肿瘤及正常卵巢组织的表达情况,探讨其与卵巢癌发生、发展的相关性及意义。方法应用免疫组化 SP 法和 RT -PCR 技术检测25例正常卵巢、25例良性卵巢肿瘤及34例上皮性卵巢癌组织中 ATG5、Beclin1、P53蛋白及 mRNA 的表达,并结合临床病理因素进行分析。结果①ATG5、Beclin1的表达主要在细胞质内,P53主要表达在胞核内。ATG5、Beclin1在正常卵巢组织、良性卵巢肿瘤组织及卵巢癌组织中的表达阳性率两两比较,差异均有统计学意义(P <0.05)。P53在卵巢癌组织中的表达阳性率与良性卵巢组织、正常卵巢组织相比,差异有统计学意义(P <0.01),而良性卵巢组织与正常卵巢组织表达阳性率比较,差异无统计学意义(P >0.05)。不同临床分期、病理分期的卵巢癌组织中 ATG5、Beclin1及 P53的表达比较,差异均有统计学意义(P <0.05)。Spearman 相关性分析显示,上皮性卵巢癌组织中 ATG5与 Beclin1蛋白表达呈正相关, ATG5与 P53、Beclin1与 P53蛋白表达均呈负相关(P <0.05)。②经 RT -PCR 检测,上皮性卵巢癌组织中 ATG5、Beclin1的表达低于良性卵巢肿瘤组织及正常卵巢组织,良性卵巢组织低于正常卵巢组织,差异均具有统计学意义(P <0.05)。上皮性卵巢癌组织中 P53的表达高于良性卵巢组织及正常组织(P <0.05),但良性卵巢组织与正常卵巢组织相比,差异无统计学意义(P >0.05)。结论上皮性卵巢癌的发病、进展可能与自噬相关基因 Beclin1和 ATG5自噬活性的抑制有关;自噬相关基因 ATG5、Beclin1与凋亡相关基因 P53在上皮性卵巢癌的发生发展中起拮抗作用,对其联合检测有利于了解和判断上皮性卵巢癌的进展程度、预后情况及指导治疗。%Objective To detect the expression of autophagy related gene ATG5,Beclin1 and apoptosis -related gene P53 in epithelial ovarian cancer,benign ovarian tumor and normal ovarian,and investigate their correlation and significance in the oc-currence and development of ovarian cancer.Methods Immunohistochemistry SP and RT -PCR methods were used to detect the expression level of ATG5,Beclin1 and P53 protein and mRNA in 25 cases of normal ovarian,25 cases of benign ovarian tumor and 34 cases of epithelial ovarian cancer tissue.And it was analyzed in combination with clinical pathological factors.Re-sults ①ATG5,Beclin1 mainly expressed in the cytoplasm,P53 mainly expressed in the nucleus.The positive expression rates of ATG5,Beclin1 in normal ovarian tissue,benign ovarian tumor and ovarian carcinoma were statistically different (P <0.05). The positive expression rates of P53 in ovarian cancer was different from benign ovarian tissue,normal ovarian tissue (P <0.01),but the expression rates between benign ovarian tissue and normal ovarian tissue were not statistically different (P >0.05).The expression rates of ATG5,Beclin1 and P53 were different in ovarian cancer tissues with different clinical and patholog-ical staging (P <0.05).Spearman correlation analysis showed that the expression of protein Beclin1 and ATG5 in epithelial ovarian carcinoma had a positive correlation;ATG5 and P53,Beclin1 and P53 protein expression had a negative correlation (P <0.05).②By RT -PCR,the positive expression rates of ATG5,Beclin1 in epithelial ovarian cancer tissues were lower than benign ovarian tumors and normal ovarian tissue;the positive expression rates in benign ovarian tissue were lower than normal ovarian tissue,and the differences were statistically significant (P <0.05).The expression level of P53 in epithelial ovarian cancer tissues was higher than benign ovarian tissue and normal tissue (P <0.05),but there was no difference between benign ovarian tissue and normal o-varian tissue (P >0.05).Conclusion The occurrence and development of epithelial ovarian cancer may be associated with the in-hibition of the activity of autophagy -related genes Beclin1 and ATG5.Autophagy -related genes ATG5,Beclin1 and apoptosis -re-lated genes P53 may play an antagonistic role in the development of epithelial ovarian cancer. Combined detection is helpful to understand and judge the degree of progression of epithelial ovarian cancer,prognosis and guide clinical treatment.
