17β-雌二醇在体外诱导人胆管上皮细胞增殖的研究

         

摘要

目的:研究利用雌激素诱导人胆管上皮细胞的增殖作用,并探讨其临床作用.方法:用17β-雌二醇作用于人胆管上皮细胞,CCK8方法检测17β-雌二醇对人胆管上皮细胞生长的影响,Westernblot胆管上皮细胞的雌激素受体(ER-α,ER-β)表达水平,并通过ELISA 检测上述两种培养条件下胆管上皮细胞培养上清中表皮细胞生长因子(epidermal growth factor,EGF)的分泌水平.结果:17β-雌二醇可以促进人胆管上皮细胞增殖,雌二醇组的人胆管上皮细胞ER-α,ER-β表达水平明显高于对照组(P<0.05),用ELISA 检测两组的上清中表皮细胞生长因子(epidermal growth factor,EGF)无显著性差异(P>0.05).结论:17β-雌二醇可以诱导人胆管上皮细胞增殖,主要通过胆管上皮细胞的雌激素受体表达水平上升,而不是表皮细胞生长因子的通路来实现的.%Objective: To study the use of estrogen-induced proliferation in human normal biliary epithelial cells and to explore its clinical role. Method: 17β-estradiol was used in human normal biliary epithelial cells , CCK-8 was employed to observe the influence on the biological characteristic of human normal biliary epithelial cells , expression of the estrogen receptor ( ER-α and ER-β ) examined by westernblot, and the culture supernatant of epidermal growth factor ( epidermal growth factor, EGF ) secretion levels by ELISA in HBECs. Result: 17β -estradiol can promote HBECs proliferation, estradiol group of people HBECs ER-alpha, ER-β expression level was significantly higher than that in the control group ( P<0. 05 ). Supernatant of epidermal growth factor ( epidermal growth factor, EGF ) were detected by ELISA. There's no significant difference ( P>0. 05 ). Conclusion: 17β-estradiol induced HBECs proliferation, mainly through the HBECs of estrogen receptor expression level rise, rather than the passage of the epidermal growth factor to achieve.
