益智仁提取物原儿茶酸对大鼠局灶性脑缺血再灌注损伤的保护作用

             

摘要

Objective To study the protective effect of protocatechuic acid (PCA) on focal cerebral isch-emic-reperfusin injury. Methods Thirty-six healthy male SD rats were randomly divided into three groups, each with 12 rats: sham operation group (group S), ischemia-reperfusion injury group (group IR) and protocatechuic acid group (group PCA). Focal cerebral ischemia-reperfusion injury was induced by right middle cerebral artery occlusion (MCAO) followed by 24 h reperfusion. In group PCA, rats were administrated PCA mtraperitoneally at 25 mg/kg once a day for 7 consecutive days before MCAO. The neurologic function was evaluated at 24 h of reperfusion. Then the rats were sacrificed, and the brains were removed for determination of infarct volume percentage, NF-κB-p65 and Notchl expressed in ischemic penumbra by PT-PCR and Western blot. Results Compared with group IR, the infarct volume percentage, NF-κB-p65 and Notchl expression were significantly lower in group PCA, while the neurologic function score was significantly higher (P<0.05). Conclusion PCA can reduce focal cerebral ischemia-reperfusion injury by inhibiting the expression of NF-KB-p65 and Notch in rats.%目的 应用SD大鼠局灶性脑缺血模型,研究原儿茶酸对脑缺血性再灌注损伤的保护作用.方法 健康雄性成年SD大鼠36只,随机分为三组(n=12):假手术组(S组)、脑缺血再灌注组(IR组)和原儿茶酸处理组(PCA组).采用大脑中动脉阻断法制备大鼠局灶性脑缺血再灌注模型.大鼠腹腔注射PCA,25mg/kg、1次/d、连续7d.PCA组于预处理结束后24 h,制备局灶性脑缺血再灌注模型.再灌注24 h后,行神经功能评分,随后处死大鼠,取脑组织,测定脑梗死体积百分比,采用PT-PCR法及Western blot测定缺血半暗带NF-κB-p65及Notch1的表达.结果 与IR组比较,PCA组脑梗死体积百分比降低,神经功能评分升高,脑组织缺血区半暗带NF-κB-p65及Notch1表达下调(P<0.05).结论 PCA预处理可能通过抑制脑组织NF-κB-p65及Notch1的表达、减轻了大鼠局灶性脑缺血再灌注的损伤.
