鲜烤烟叶酵母菌的分离鉴定及其碳代谢特征

         

摘要

为探明Biolog微生物自动鉴定系统在烤烟酵母菌鉴定中的应用,为鲜烤烟叶上微生物分析奠定理论基础,以K326烤后烟叶为材料,采用涂布培养法对鲜烤烟叶表面可培养酵母菌进行分离,菌落观察和镜检,采用Biolog微生物鉴定系统对分离的酵母菌进行鉴定;采用鉴定过程中生化反应进行酵母菌的碳代谢特征分析.结果表明:鲜烤烟叶上共分离得到7株酵母菌,其中,锁掷酵母菌(Sporidiobolus johnsonii)4株,白吉利毛孢子菌(Trichosporon beigelii)2株,橙黄红酵母菌(Rhodotorula aurantiaca)1株.3种酵母菌在碳源代谢和同化能力上存在差异,糊精是3种酵母菌能同时利用的唯一碳源.%In order to analyze the utilization of Biolog Automated Microbes Identification System in identifying yeast that hosted from fresh flue-cured tobacco leaves,and also to get some theoretical basis for studying tobacco leaf microorganism,tobacco yeasts in fresh-cured tobacco leaves (cv.K326) were isolated by spread plate method.They were checked by morphology study on the plates and with the help of a microscopy,then identified by Biolog Automated Microbes Identification System,and also analyzed for carbon utilization in the identification plate.Results:Seven yeast isolates were obtained and identified,including four Sporidiobolus johnsonii species,two Trichosporon beigelii species,and one Rhodotorula aurantiaca species.Three species had many differences in carbon utilization and assimilation.Dextrin is the only carbon that all three yeasts can use for all three species.
