帕瑞昔布钠不同方式预先给药对神经病理性疼痛大鼠行为学的影响

     

摘要

目的:通过鞘内、腹腔、损伤神经周浸润3种不同给药方式,探讨帕瑞昔布钠预先给药对L5脊神经结扎大鼠神经病理性疼痛行为学的影响。方法成年SD雄性大鼠30只,随机分5组:假手术组( SC组)、模型组( SNL组)、帕瑞昔布钠鞘内注射400μg组( IT组)、腹腔8 mg/kg组( IP组)和神经周围浸润400μg组( PI组),每组6只。各给药组大鼠均在神经结扎前30 min经上述不同途径给予帕瑞昔布钠进行预先给药。监测术前1 d,术后3、7、10、14、21 d各组大鼠双侧后肢的50%机械缩足反应阈值(50%PMWT)和热刺激缩足反应潜伏期( PTWL)。结果单次给予鞘内400μg、腹腔8 mg/kg、神经周浸润400μg帕瑞昔布钠预先给药均可有效抑制大鼠神经结扎损伤后神经病理性疼痛的形成,与SNL组相比,3个给药组大鼠损伤侧(左)各时间点的50%PMWT、PTWL均明显升高(P<0.05);其中IT组升高最为明显,分别与IP组及PI组相比,各值差异均有统计学意义(P<0.05),而IP与PI两组之间的差异无统计学意义( P>0.05)。除SC组外,其他各组非损伤侧(右)在造模后也出现了50%PMWT、PTWL不同程度的短期降低,鞘内给予400μg帕瑞昔布钠可效抑制其PTWL的降低程度。结论3种给药方式的帕瑞昔布钠预先给药对脊神经结扎后大鼠热痛敏和机械痛敏的形成均有一定程度的抑制作用,其中以鞘内预先给药的效果最佳,神经周围浸润给药与腹腔给药效果相似。%Objective To compare the behavioral effects of parecoxib pretreatment on the rats with Neuropathic pain by different administration pathways , including intrathecal and intraperitoneal injection and perineural infiltration . Methods Thirty adult male rats were randomly divided into five groups: SC controlled group ( Group SC ) , L5 spinal nerve ligation group (Group SNL), parecoxib intrathecal injection group (Group IT), parecoxib intraperitoneal injection group (Group IP), perineural parecoxib infiltration group (Group PI).Rats in treatment groups were given with parecoxib pretreatment.Fifty percent paw mechanical withdrawal threshold (50%PMWT) and paw thermal withdrawal latency (PT-WL) were measured on the 1st pre-operative and 3rd, 7th, 10th, 14th, 21st post-operative day.Results The post-op-erative left 50%PMWT in Group SNL and the three treatment groups were significantly lower than those in Group SC ( P<0.05);meanwhile, the post-operative left 50%PMWT in the three treatment groups were significantly higher than those in Group SNL (P<0.05).The post-operative left 50%PMWT in Group IT were significantly higher than those in Group IP and Group PI ( P<0.05 ) , and there were no difference between left 50%PMWT in Group IP and Group PI ( P>0.05).Compared with Group SC, noticed reduced right 50%PMWT in the other four group were also observed , which was partially inhibited by intrathecal pretreatment of parecoxib .Conclusion All of the parecoxib pretreatment by three different administration pathways can alleviate the mechanical and thermal pain after spinal nerve ligation in rats .Intrathe-cal administration provides the best efficacy , while the other two administration pathways provides similar results .
