目的 观察复方贞术调脂胶囊(FTZ) 对体外培养破骨细胞的影响,探求相关作用机制.方法 采用体外分离培养破骨细胞,随机分为control组、Alendronate组、FTZ-H 组、FTZ-M 组和FTZ-L 组,各组大鼠予含药血清、M-CSF 和RANKL 细胞刺激因子诱导培养8 d,电子显微镜下观察破骨细胞数量和骨陷窝面积、RT-PCR 法检测Trap、MMP-9、NFATc1 mRNA 的表达.结果 经Trap染色,均可在5倍镜下见大小不一、细胞核数不一的多核破骨细胞,与control组相比,Alendronate组、FTZ-H 组和FTZ-M 组的破骨细胞(OC) 数量减少(P ＜ 0. 05,P ＜ 0. 01) ,FTZ 组间比较,FTZ-H 组较FTZ-L 组破骨细胞数量显著降低(P ＜ 0. 05);Alendronate组、FTZ-H 组和FTZ-M 组的骨陷窝面积较control组显著减少(P ＜ 0. 05) ,FTZ 组间比较,FTZ-H 组骨陷窝面积较FTZ-L 组减少,但差异无统计学意义(P ＞ 0. 05) .与control组比较,Alendronate组、FTZ-H 组Trap、MMP-9、NFATc1 mRNA 表达均显著降低(P ＜ 0. 01) .结论 FTZ-H 和FTZ-M 浓度具有抑制破骨细胞分化和骨吸收活性的作用,其中FTZ-H 组与Alendronate组无异.%Objective To investigate the effect of FTZ on the bone absorption ability of osteoclasts in vitro. Methods Osteoclasts were separated from C57 B6/ J of 4-6w mice and cultured. They were randomly divided into five groups, control, Alendronate, FTZ-H, FTZ-M and FTZ-L groups. After 8 days of cultivation, the number of osteoclast and bone absorptive lacunae were counted by electron microscope. Trap, MMP-9 and NFATc1 mRNA were assessed by RT-PCR. Results Compared with the control group, the number of osteoclasts and the area of bone absorptive lacunae were significantly reduced Alendronate, FTZ-H and FTZ-M groups (P ＜ 0. 01, P ＜ 0. 05). Compared with FTZ-L group, significant reduction in the number of osteoclasts was observed in FTZ-H group. There is no significant difference between FTZ-L and FTZ-H in the area of bone absorptive lacunae (P ＞ 0. 05). The gene expressions of Trap, MMP-9 and NFATc1 were significantly lower in Alendronate and FTZ-H groups than that in control group (P ＜ 0. 01). Conclusion FTZ-H and FTZ-M inhibit osteoblast differentiation and bone absorbing activity, but there is no significant difference between FTZ-H and FTZ-M.
