升阳益胃汤不同提取方法对其抗炎效应的影响

         

摘要

Objective To evaluate the effects of different extraction process of Shengyang Yiwei decoction on anti-inflammation in intro and vivo. Methods Shengyang Yiwei decoction was extracted by three process, which was respectively marked in M1, M2 and M3. Effects of different extraction process on dissolution of Shengyang Yiwei decoction were evaluated by high performance liquid chromatography (HPLC). Moreover, fifty KM mice were arranged into normal, model, M1, M2, M3 groups by body weight, and the body weight was recorded every other week. After drug treatment for two weeks, besides of normal group, other mice were treated with 0.04 m L xylene. Four hours later, mice were sacrificed by cervical vertebra separation, and then, ear weight difference, ear thickness difference, thymus weight and spleen weight were recorded, meanwhile, ear swelling degree and visceral index were calculated. Furthermore, effects of different extraction process on HBZY-1 cell proliferation were detected by MTT. In addition, effects of different extraction process different extraction process on macrophage infiltration induced by AGEs (advanced glycation end products) was detected by transwell. Results (1) Characteristic peak area of M3 group was the highest. (2) Ear weight difference, ear thickness difference and ear swelling degree were significantly increased in model group, compared with normal group; after drug treatment, ear weight difference, ear thickness difference and ear swelling degree were significantly decreased, but body weight was markedly increased in M1, M2 and M3 group, compared with model group. (3) When the dosage of administration was less than or equal to 10 mg herb/m L, there was no significant inhibition between HBZY-1 cell and drugs. (4) Macrophage infiltration degree was markedly reduced in M1, M2 and M3 group, compared with model group. Furthermore, the anti-infiltration ability from high to low was M3, M2 and M1. (5) There was negative relation between characteristic peak area and macrophage infiltration degree.Conclusion Different extraction process could inhibit ear swelling of mice induced by xylene, but there was no significant difference between them. In addition, different extraction process could inhibit macrophage infiltration induced by AGEs, and the anti-infiltration ability from high to low was M3, M2 and M1.Decoction of all herbs together was conducive to dissolution of anti-inflammation components, and then increasing anti-inflammation activity of Shengyang Yiwei decoction.%目的 比较升阳益胃汤不同提取方法对其抗炎作用的影响.方法 采用不同提取方法对升阳益胃汤进行提取,提取方法分别标记为M1、M2、M3.高效液相法检测不同提取方法对升阳益胃汤成分溶出的影响.昆明种小鼠50只,按照体重随机分为正常组、模型组、M1组、M2组、M3组.给药14天后,除正常组小鼠外,其余各组均采用二甲苯0.04 m L/只的剂量造模,正常组小鼠左右耳及造模小鼠左耳均涂抹同等剂量蒸馏水.4小时后将小鼠颈椎断离处死,打耳片后记录耳重量差、耳厚度差,计算耳肿胀度;记录胸腺、脾脏重量,计算其脏器指数.MTT法检测不同提取方法对HBZY-1细胞增殖的影响.Transwell实验检测不同提取方法的抗炎作用.结果(1)M3组特征峰面积最大.(2)与正常组相比,模型组小鼠耳重量差、耳厚度差、耳肿胀度均显著增加;与模型组相比,给药后M1、M2、M3组小鼠体重显著升高,耳重量差、耳厚度差、耳肿胀度均显著降低.(3)M1、M2、M3组给药剂量≤10 mg生药/m L时,对HBZY-1细胞的生长无明显抑制作用.(4)与模型组相比,M1、M2、M3组巨噬细胞浸润程度明显降低,且抗巨噬细胞浸润作用为M3> M2> M1.(5)特征峰面积与巨噬细胞浸润度之间存在显著负相关.结论 不同提取方法均可抑制二甲苯所致小鼠耳肿胀,且抑制作用无显著性差异;不同提取方法均可以抑制巨噬细胞浸润,且M3的作用明显优于M1和M2.综合体、内外抗炎作用研究发现,全方合提可增加升阳益胃汤抗炎活性物质溶出,增加其抗炎活性.
