不同浓度虫茶对MCF-7人乳腺癌细胞的体外抗癌效果

     

摘要

对购买的一种虫茶进行MCF-7(人乳腺癌细胞)体外抗癌效果评价。通过MTT试验、DAPI荧光染色分析、RT-PCR和western blot分析,研究其抗癌效果。研究结果表明,200μg/mL浓度的虫茶(78%)对MCF-7人乳腺癌细胞生长相对最强的抑制效果。DAPI染色分析显示,与100和50μg/mL虫茶相比,使用200μg/mL浓度的虫茶,对MCF-7细胞有较强的诱其凋亡的能力,Bax, caspase-3和caspase-9的mRNA和蛋白质表达得到增强, Bcl-2表达减弱。综合分析得出,一定浓度的虫茶具有良好的抗癌预防效果。%One sandy tea product was purchased for the evaluation of in vitro anticancer effect in MCF-7 human breast adenocarcinoma cells. In MTT assay, apoptosis analysis by DAPI staining and RT-PCR, western blot gene expressions were employed to check their anticancer effect.At the concentration of 200μg/mL, sandy tea (78%) showed the best inhibitory effect on the growth of MCF-7 human breast adenocarcinoma cells. DAPI staining showed that 200μg/mL of sandy tea induced apoptosis on MCF-7 cells. Sandy tea (200μg/mL) significantly induced apoptosis, up-regulated mRNA and protein expressions of Bax, caspase-3 and caspase-9, and down-regulated Bcl-2 expression, compared with 100 and 50μg/mL of sandy tea. These results suggest that sandy tea showed good cancer preventive effect.
