反相硅胶纯化阿克苏“次等枣”环磷酸腺苷

     

摘要

采用反相硅胶对红枣环磷酸腺苷提取物进行纯化.以吸附率和解析率为指标,确定反相硅胶纯化红枣环磷酸腺苷的最佳工艺参数为:上样液流速1 mL/min、上样液浓度70 μg/mL、上样液pH 6,洗脱液为体积分数60%的甲醇、洗脱液流速3 mL/min,在此条件下环磷酸腺苷的纯度由0.098%提高到44.69%.%Reversed phase silica gel column was applied for purifying Cyclic adenosine phosphate (cAMP) with purity as index.The optimal purification parameters were determined with adsorption and desorption rate as indicators,and the results showed that the sample fluid velocity was 1mL/min,the sample concentration was 70μg/mL,pH 6,the flow velocity 3mL/min with 60% methanol as eluent.Under these conditions,the purity of cAMP was increased from 0.098% to 44.69%.
