Objective:Using response surface methodology to optimize extraction technology of total alka-loids from Fritillaria pallidiflorae bulbus. Method : Based on single factor test, ethanol concentration, effects of extraction time and liquid-solid ratio on yield of total alkaloids from Fritillaria pallidiflorae bul-bus was investigated with a three-factor Box-Behnken center-united design. The comprehensive score of the total alkaloid content and extract yield as response value was optimized by response surface methodol-ogy. Result:Optimized extraction technology conditions was as following:43. 14 times of 69. 78% ethanol , pH 2. 0, extract 1 times, each time 1. 86 h. According to actual situation in production, optimum ex-traction parameters were modified as 40 times of 70% ethanol , pH 2. 0, extract 1 times, each time 1. 5 h, the total alkaloid content reached 11. 04%. The total alkaloids of Fritillaria extract rate reached 13. 75%, the comprehensive score of 98. 90 under these conditions. Conclusion:actual value which was in consistency with the theoretic value. This modified extraction parameters were credible and reliable with some practical value, and can be used for the extraction of alkaloids from Fritillaria.%目的::采用响应曲面法( Box-Behnken)优化伊贝母中总生物碱的提取工艺。方法:在单因素试验基础上,选取乙醇体积分数、提取时间和液料比3个因素进行Box-Benhnken中心组合设计,以总生物碱含量和浸膏率的综合评分为响应值,利用响应曲面法对其提取工艺参数进行优化。结果:优选出的伊贝母总生物碱最佳提取工艺为加43.14倍pH 2的69.78%乙醇提取1次,每次1.86 h,考虑到生产实际操作,故将实验条件确定为加40倍pH 2的70%乙醇提取1次,每次1.5 h。在此条件下,伊贝母总生物碱含量达到11.04%,浸膏率达到13.75%,综合评分为98.90。结论:实测值与响应曲面法回归模型的预测值相近,与模型拟合良好,优化的提取工艺准确可靠,具有一定的实用价值,可用于伊贝母总生物碱的提取工艺。
