弥漫性脑创伤后Bcl-2家族与线粒体途径细胞凋亡的相关性分析

         

摘要

Objective To investigate the effectof Bc12 fan ily in m itochondrk approach apoptosis follow ing teamatic biain injuy M ethods The in peoved m odel of m oderate cbsed biain in jury w as established in rate by using of the m ethod found by M armarou .After traumatic brain injuy ,cytrc bax bc12 and counthy of apoptosis neural cell in the cortex of rate was observed dynan ically w ith the m ethod of in m unohistochan ry and TUN EL .Resulte The in m unoreaction of Cyte w as related to the quantity of TUN EL positive cells from th to 24 h (r=0 .958 ,P<0 .05) ;The ratio ofBax and Bc12 was related to the in m unoieaction of Cyte from 1h to 24h (r=0.996,P<0.01),but the peak of the in m unoreaction of Cyte (24h) was earlier than that of the ratio of Bax and Bc12 (48h) ;The ratio ofBax and Bc12 was related to the quantily of TUNEL positive cells from 1h to 72h ( r=0 .925,P<0 .01) after traumatic brain in jury .Conclusion The ratio ofBax andBc12 was inportantin regulation ofCyte release and in modulation of apoptisis gene expression after Cyte release%目的 探讨脑创伤后Bcl-2家族在线粒体途径细胞凋亡中的作用.方法 采用改进的Marmarou's法建立大鼠中度闭合性弥漫性颅脑损伤模型.应用免疫组化,TUNEL法对大鼠伤后皮质细胞色素 c、Bax、Bcl-2表达及凋亡细胞数进行动态观察.结果 伤后1 h至24 h Cytc表达与TUNEL阳性细胞数密切相关(r=0.958,P<0.05);伤后1 h至24 h Bax和Bcl-2二者的比例与Cytc免疫反应相关(r=0.996,P<0.01),但Cytc表达峰值(24 h)早于Bax和Bcl-2二者的比例高峰值(48 h);伤后1 h至72 h Bax和Bcl-2二者的比例与TUNEL阳性细胞数密切相关(r=0.925,P<0.01).结论 Bax和Bcl-2二者的比例对Ctyc释放的调节起重要作用,Bax和Bcl-2二者的比例在Ctyc释放后的凋亡基因的调节中可能仍发挥重要的作用.
