降压药物对高血压大鼠血管平滑肌细胞离子泵活性及分泌血管紧张素Ⅱ的影响

摘要

Objective To observe the effects of different types of antihypertensive drugs on activities of Na+ , K+-ATPase, Ca2+-ATPase and autocrine of Ang Ⅱ in vascular smooth muscle cells (VSMC) isolated from spontaneously hypertensive rats (SHR). Methods The thoracic aorta VSMC isolated from SHR were cultured. The cultured aorta VSMC were treated with different types of antihypertensive drugs: lisinopril, amlodipine, irbesartan, metoprolol and hydrochlorothiazide. The activities of ion pumps were detected by spectrophotometry and the content of Ang Ⅱ in cell-culture medium was detected by radioimmunoassay. Results The activities of Na+ , K+-ATPase and Ca2+-ATPase in aorta VSMC from SHR were significantly lower than those from Wistar-Kyoto rats (P ＜ 0.01) ;lisinopril arid irbesartan significantly increased the activities of the two ion pumps in aorta VSMC from SHR (P ＜ 0.01). Amlodipine only increased the activities of Na+ ,K+-ATPase,but metoprolol and hydrochlorothiazide had no effects on them. Lisinopril and amlodipine attenuated the content of Ang Ⅱ in aorta VSMC from SHR (P ＜ 0.05), but irbesartan increased it (P ＜ 0.05). Conclusion Lisinopril,amlodipine and irbesartan can increase the activities of Na+ , K+-ATPase and(or) Ca2+-ATPase and influence the autocrine of Ang Ⅱ in aorta VSMC from SHR.%目的 观察不同类型的降压药物对自发性高血压大鼠(SHR)动脉血管平滑肌细胞钠泵、钙泵活性及分泌血管紧张素Ⅱ(Ang Ⅱ)的影响.方法 采用组织块种植法培养大鼠胸主动脉血管平滑肌细胞24 h后,分为WKY大鼠组、SHR组、赖诺普利组、氨氯地平组、伊贝沙坦组、氢氯噻嗪组和美托洛尔组.分别用不同药物对SHR动脉血管平滑肌细胞进行干预.采用生化酶学方法、放射免疫分析法分别测定细胞膜离子泵活性和Ang Ⅱ浓度.结果 与WKY大鼠组比较,SHR组大鼠钠泵和钙泵活性明显降低(P<0.01),Ang Ⅱ浓度明显升高(P< 0.05);与SHR组比较,赖诺普利组、伊贝沙坦组钠泵和钙泵活性明显增高(P<0.05,P<0.01);氨氯地平组钠泵活性明显增高(P< 0.01);氢氯噻嗪组和美托洛尔组钠泵、钙泵活性及Ang Ⅱ浓度差异无统计学意义.与SHR组比较,赖诺普利组、氨氯地平组Ang Ⅱ浓度明显降低(P< 0.05),伊贝沙坦组Ang Ⅱ浓度明显升高(P< 0.05).结论 赖诺普利、伊贝沙坦和氨氯地平能提高SHR动脉血管平滑肌细胞膜离子泵活性,并影响其分泌Ang Ⅱ.
