Objective To investigate the expression and significance of Cys C in alcoholic liver disease in rats. Methods Rat model of alcoholic liver disease were established by alcohol administration. Rats were killed at the end of 12 weeks. Liver indexes of the rats were calculated. Levels of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were measured. The pathological change of liver was observed by H-E staining. The protein expression of Cys C was detected by immunohistochemistry. The mRNA expression of Cys C was determined by RT-PCR. Results Compared with control group, liver indexes in model group increased (P < 0. 01 ). H-E staining showed hepatocytes swelling, macro or microvesicular steatosis and inflammatory cells infiltration in cytoplasm in the model group. The levels of serum AST and ALT increased significantly in model group (P < 0.05) compared with control group. Protein and mRNA expression of Cys C in the liver determined by immunohistochemistry and RT-PCR increased significantly in the model group (P <0.01 ). Conclusion Cys C may be involved in the genesis and development of ALD.%目的 探讨胱蛋白酶抑制剂C(Cystatin C,Cys C)在大鼠酒精性肝病中的表达及意义.方法 酒精灌胃法制备大鼠酒精性肝病模型.12周末处死大鼠,测定大鼠肝指数,检测血清天门冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)含量,HE染色观察肝脏病理学改变,免疫组化SAB法比较正常组和模型组Cys C蛋白的表达,RT-PCR法比较两组肝组织Cys C mRNA的表达变化.结果 模型组肝指数较正常组显著升高(P<0.01);HE染色观察模型组肝细胞肿胀,包浆内可见大小不等的脂肪空泡及炎性细胞浸润;模型组血清ALT和AST较正常组明显升高(P<0.05);免疫组化及RT-PCR显示,肝组织中Cys C蛋白及mRNA表达模型组较正常组显著上调(P<0.01).结论 肝组织 Cys C 表达上调可能参与了酒精性肝病的发生、发展过程.
