Objective To evaluate the biochemical function of rat nature three-dimensional acellular pancreatic bioscaffold (3D-APB).Methods The fresh pancreas from 10 rats were perfused to produce 3D-APB.The biocompatibility of 3D-APB were evaluated.The experiment was divided into 4 groups based on 4 kinds of three-dimensional media for AR42J culture:blank control group,extracellular matrix (ECM) group,Poly-lactic-co-glycolic-acid (PLGA) group and 3D-APB group.We compared the proliferation and differentiation of AR42J pancreatic acinar cell at 3 days,5 days,7 days,10 days after seeding among 4 groups.Results The 3D-APB represents a biocompatible scaffold capable of integrating within host tissue.The proliferation rate of AR42J cell on 3D-APB is higher (P =0.011)while the apoptosis rate is lower (P =0.017) than on other 3 medias with significant difference,respectively.The protein expression of pancreatic duodenal homeodomain containing transcription factor (PDX-1)and pancreatic exocrine transcription factor (PTF-1) in 3D-APB were higher than other 3 groups with statistical significance (PPDX-1 =0.013,PPTr-1 =0.012).Also,the gene expression of PDX-1 and PTF-1 mRNA in 3D-APB were higher than other 3 groups with statistical significance (PPDx-1 =0.013,PPTF-1 =0.012).Conclusion 3D-APB can promote cell proliferation and differentiation which is benefit for regeneration medicine utility compared with other kinds of chemosynthesis and native component scaffolds.%目的 体外评估大鼠天然全胰腺脱细胞3D支架(3D-APB)促进种植细胞增殖和分化的功能.方法 利用改良灌注技术制备大鼠3D-APB,随即将实验分为4组,评估培养后第3、5、7、10天细胞的增殖活性及细胞分化功能.结果 3D-APB组细胞增殖率高于其他3组(P=0.011),而凋亡率低于其余3组(P=0.017),且差异有统计学意义;3D-APB组胰十二指肠同源结构域转录因子(PDX-1)和胰腺外分泌转录因子(PTF-1)蛋白表达均高于其余3组,且差异有统计学意义(PPDX-1 =0.013,PPTF-1=0.012);3D-APB组PDX-1和PTF-1基因表达含量均高于其余3组,且差异有统计学意义(PPDX-1 =0.013,PPTF-1=0.012).结论 3D-APB更有利于促进细胞增殖及分化,更适合应用于再生医学领域.
