白藜芦醇对急性心肌梗死大鼠心肌保护及Akt/JNK3/caspase-3信号通路的影响

     

摘要

目的 探究白藜芦醇(Res)对急性心肌梗死(AMI)大鼠氧化应激及对p-Akt、p-JNK3、caspase-3的影响.方法 将大鼠分为假手术组、模型组、法舒地尔组、Res低剂量组、Res中剂量组、Res高剂量组;检测血清中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、肌钙蛋白I(TnI)、超氧化物歧化酶(SOD)、丙二醛(MDA)的活性及白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量;TTC染色检测心肌梗死面积;HE染色检测心脏组织病理学变化;采用TUNEL检测心肌细胞凋亡;蛋白免疫印迹检测心肌组织中p-Akt、p-JNK3、caspase-3表达情况.结果HE染色结果表明,模型组心肌细胞排列紊乱,心肌纤维肿胀、断裂;与模型组相比,法舒地尔组和Res组心肌细胞和肌纤维排列相对整齐,炎性细胞浸润情况减轻,且心肌梗死面积和细胞凋亡指数显著低于模型组(P<0.05);与模型组相比,法舒地尔组和Res组SOD的活性显著增高(P<0.05),血清中CK、CK-MB、TnI、MDA的活性及IL-1β、IL-6、TNF-α 的含量显著低于模型组(P<0.05),且法舒地尔组和Res组心脏组织中p-Akt表达水平显著高于模型组(P<0.05),p-JNK3、caspase-3表达水平显著低于模型组(P<0.05);与法舒地尔组相比,Res高剂量组血清中CK、CK-MB、TnI、MDA的活性,血清中IL-1β、IL-6、TNF-α 的含量,心肌梗死面积和细胞凋亡指数,以及心肌组织中p-Akt、p-JNK3、caspase-3表达水平等,差异均无统计学意义(P>0.05).结论 Res对AMI大鼠心肌具有保护作用,推测这一作用是通过调控Akt / JNK3 / caspase-3信号通路从而抑制心肌细胞凋亡.%Objective To investigate the influences of Resveratrol (Res) on oxidative stress and p-Akt, p-JNK3 and Caspase-3 in rats with acute myocardial infarction (AMI). Methods The rats were divided into sham operation group, model group and Fasudil group, Res low dose group, Res medium dose group, Res high dose group; the activities of serum SOD, CK-MB, CK, MDA and the contents of IL-1β, IL-6 and TNF-α in were detected; TTC staining was used to detect the area of myocardial infarction; the changes of cardiac histopathology were detected by HE staining; TUNEL was used to detect the apoptosis of myocardial cells; the expressions of p-Akt, p-JNK3 and Caspase-3 in myocardial tissue was detected by Western blot. Results The results of HE staining showed that the myocardial cells in the model group were arranged in disorder, the myocardial fibers were swollen and fractured;compared with the model group, myocardial cells and muscle fibers in Fasudil group and Res group were relatively neat, the infiltration of inflammatory cells was reduced; the activity of SOD in Fasudil group and Res group were significantly higher than that in model group (P<0.05), the activities of serum CK-MB, CK, MDA and the contents of IL-1β, IL-6, TNF-α in Fasudil group and Res group were significantly lower than those in the model group (P<0.05); Fasudil group and Res group, the myocardial infarct size and apoptosis index in Fasudil group and Res group were significantly lower than those in the model group (P<0.05); the expression level of p-Akt in cardiac tissue of fasudil group and Res group was significantly higher than those of the model group (P<0.05); the expression levels of P-JNK3 and Caspase-3 in cardiac tissue of fasudil group and Res group was significantly lower than those of the model group (P<0.05); the activities of serum CK-MB, CK, MDA, the contents of IL-1β, IL-6, TNF-α, the myocardial infarct size and apoptosis index, the expression levels of p-Akt, P-JNK3 and Caspase-3 in the Res high dose group were not significantly different with those in the Fasudil group (P>0.05). Conclusion Res has protective effect on the myocardium of AMI rats, it is presumed that this effect is achieved by regulating the Akt/JNK3/caspase-3 signaling pathway to inhibit cardiomyocyte apoptosis, it provides a certain theoretical basis for the clinical treatment of AMI.
