血府逐瘀汤诱导内皮细胞迁移中Ⅳ型胶原酶的作用研究

摘要

目的:探讨血府逐瘀汤影响Ⅳ型胶原酶诱导内皮细胞迁移的作用.方法:运用血清药理学方法,以1.25％、2.5％和5％浓度的血府逐瘀汤含药血清和空白血清培养人脐静脉内皮细胞ECV304,采用Boyden小室迁移法检测药物对内皮细胞迁移的影响,并通过酶联免疫法和RT-PCR检测明胶酶A(MMP-2)和明胶酶B(MMP-9)分泌和表达的情况.结果:1.25％浓度含药血清对上述指标无影响,2.5％和5％浓度含药血清可显著提高内皮细胞迁移、MMP-2和MMP-9转录及细胞培养上清液中MMP-2和MMP-9水平.结论:血府逐瘀汤通过提高Ⅳ型胶原酶的表达进而诱导内皮细胞迁移,提示药物促血管新生功能可能与该作用相关.%Objective: To investigate the effect of type IV collagenase on endothelial cell migration induced by Xuefu Zhuyu Decoction(XFZYD). Methods; Serum pharmacology technique was adopted. Endothelial cells ECV304 were treated with blank serum or XFZYD containing serum( XFZYD-CS) at final concentrations of 1.25% , 2.5% and 5%. Boyden chamber assay was performed to test cell migration function. ELISA and RT-PCR was used to estimate MMP-2 and MMP-9 level in cell supernatant and their transcription. Results; Final concentration of 2. 5% and 5% XFZYD-CS promoted cell migration as well as MMP-2 and MMP-9 level and their transcription remarkably. Conclusion; XFZYD could induce ECV304 migration through high expression of MMP-2 and MMP-9. This might account for partial mechanism of angiogenesis effect induced by XFZYD.
