大鼠C6胶质瘤模型建立及7T动物磁共振表现实验研究

         

摘要

Purpose: To establish the model of rat C6 glioma and study their features with 7T animal MRI, including conventional MR imaging, DWI, MRS and PWI. And to optimize the MR imaging sequences and parameters. Methods: Forty SD rats with body weight 200g were chosen and implanted C6 glioma cells at right caudate nucleus of the brain through stereotaxic apparatus to establish rat brain glioma models. The location of needling was about lmm lateral to biparietal suture and 3mm right of arcuate suture. The depth of needling was 5mm and back lmm. The total needling liquid was l0ul with cell density of 2 x 10V μl. The total needling time was about 10 minutes. The needle was drawn after 5 minutes . Seven days after glioma cells implanting, all rats were examined with MRI to choose the rat glioma of the same size and location for study. The chosen 30 rats were examined with conventional MRI, DWI, MRS, and PWI. After MRI examination, the rats were sampled for electronic microscopy and pathology exam. Results: After 7 days development of the tumor, the success rate was 100% . The average volume of glioma was 0.15cm3. C6 rat brain gliomas were proved by pathology and microscopy. The golimas were with long Ti and long T2 signal on routine MRI, high signal on DWI. The average rADC value was 0. 70 x 10 3m/s. The baseline of MRS was stable and the ratio of area under peak of Cho/Cr, NAA/Cr, Lac/Cr, Lip/Cr and Cho/NAA was 1.22, 1.67, 1.70, 0.76, 0.75, respectively. The PWI curve was stable and theaverage rrCBV value was 3.73. Conclusion: The model of rat brain glioma could be established by implanting C6 glioma cells through stereotaxic apparatus. The 7T MRI images of all sequences are clear and stable, the optimal parameters of 7T MRI sequences are found successfully, so they are useful for future experiment.%目的:制备大鼠脑C6胶质瘤模型并进行7T磁共振成像(包括常规MRI、DWI、MRS、PWI),研究大鼠脑胶质瘤磁共振成像特点,优化成像序列及参数.方法:选取SD大鼠40只,体重约200g;在立体定向仪下,以矢状缝旁1mm及冠状缝右侧约3mm确定为进针点,进针深度为5mm后拔出1mm,缓慢注入浓度为2x 105个/μlC6胶质瘤细胞混悬液约10μl,注射10min,停针5min后拔出;种植后第7天进行磁共振筛选,选取大小及部位基本一致的肿瘤30只,采集常规MRI及功能磁共振成像(包括DWI、MRS、PWI);检查完毕,获取胶质瘤标本送病理学及电镜检查.结果:大鼠脑胶质瘤成瘤率100%;肿瘤平均体积约为0.15cm3,病理学及电镜检查证实为C6胶质瘤;常规MRI呈长T1长T2信号,DWI呈稍高信号,平均rADC值为0.70x 10-3m/s,MRS基线稳定,各代谢物峰下面积与肌酐比值Cho/NAA比值分别为Cho/Cr1.22,NAA/Cr1.67,Lac1.70, Lip0.76,Cho/NAA0.75.PWI曲线稳定,相对血容量rrCBV值为3.73.结论:通过立体定向仪种植胶质瘤细胞混悬液成功制备鼠脑胶质瘤模型,7T MRI各序列成像清晰稳定,成功摸索出7T MRI的各种序列参数,为后续实验奠定基础.
