薄荷脑对葛根素体外黏膜的促渗作用研究

         

摘要

目的:研究薄荷脑对葛根素体外黏膜的促渗作用.方法:选择离体牛蛙腹皮作为渗透模型,采用Franz扩散池进行体外黏膜渗透试验,高效液相色谱法测定葛根素的含量,考察单用葛根素及葛根素与薄荷脑不同比例混合对表观渗透系数(Papp)的影响.结果:不同浓度的葛根素对Papp的影响无显著性差异;配伍薄荷脑后,当薄荷脑浓度达到5 mg·mL-1时,与单用葛根素比较Papp有显著性差异,且此时葛根素与薄荷脑不同比例组间的Papp无显著性差异.结论:葛根素的黏膜渗透属于被动扩散;薄荷脑在一定浓度范围内能够促进葛根素的渗透,当浓度达到5 mg·mL-1时具有显著促渗作用,在此浓度下,葛根素浓度的增大不会影响到薄荷脑的促渗作用.%OBJECTIVE: To study the influence of menthol on mucosal permeability of puerarin in vitro. METHODS: Take the abdominal skin of rana catesbeiana as a vitro model. The effect of different concentrations of puerarin and different compatibility proportion between them were investigated separately, by using Franz diffusion cell and HPLC. The content of puerarin was determined by HPLC, and the effects of puerarin alone and different proportion of puerarin to menthol on apparent permeability coefficient were investigated. RESULTS: There was no significant difference in the effects of different concentrations of puerarin on Pspp. When mass concentration of menthol was 5 mg·mL-1 after compatibility, there was significant difference between menthol+puera-rin group and puerarin group. And at this concentration,Papp of different compatibility proportion groups showed no difference. CONCLUSION : The permeation of puerarin is considered as passive diffusion. The menthol can enhance permeation in a certain range of concentration, and gain the significant effect when the concentration of menthol was 5 mg·mL-1. And at this concentration, concentration change of puerarin will not have negative influence on permeation.
