甲泼尼松龙对多索茶碱在大鼠体内药动学的影响

摘要

Objective To study the effect of single dose methylprednisolone on the pharmacokinetics of doxofylline in rats. Methods Male rats were randomly divided into doxophylline group and combined treatment group (doxophylline+methylprednisolone).RP-HPLC method was adopted to determine the serum concentration for different time before and after administration of doxofylline.Pharmacokinetic parameters were calculated by PK Solver 2.0 pharmacokinetics soft-ware.Pharmacokinetic parameters were compared between two groups of rats. Results The two groups had statistical dif-ference in rats with AUC0~t,AUC0~∞,t1/2(P<0.05).The indexs of combined treatment group were increased than that of dox-ophylline group,Cl_obs group of combined treatment group was decreased compared with doxofylline group. Conclusion Methylprednisolone has influence on the pharmacokinetics of doxofylline in rats,will slow the body’s metabolism and e-limination.%目的：考察单次尾静脉注射甲泼尼松龙对大鼠体内多索茶碱药动学的影响。方法将12只雄性大鼠随机分成多索茶碱组及合并用药组，用反相高效液相色谱法（RP-HPLC）测定大鼠给药前后不同时间点多索茶碱的血清浓度，通过PK Solver 2.0软件拟合计算，对两组大鼠的多索茶碱主要药动学参数进行比较。结果两组大鼠多索茶碱AUC0~t、AUC0~肄、t1/2比较差异有统计学意义（P<0.05），合并用药组较单独用药组均增加；而Cl_obs合并用药组较多索茶碱组减少。结论甲泼尼松龙可能对多索茶碱在大鼠体内的药动学有影响，会减慢其体内的代谢和消除。
