METHOD FOR ASSESSING EFFECT OF ADEMETHIONINE ON ENERGY METABOLISM, PROOXIDANT-ANTIOXIDANT SYSTEM IN LIVER, MYOCARDIUM, AND BRAIN OF RATS IN DICHLORETHANE-INDUCED HEPATITIS
The method for assessing the effect of ademethionine on the energy metabolism, prooxidant-antioxidant system in liver, myocardium, and brain of the rats in dichlorethane-induced hepatitis comprises blood examination. The toxic hepatitis is induced by single administration of dichlorethane in a volume of 500 mL/kg. On the fifth day, Ademethionine is administered by gavage at a dose of 100 mg/kg. The biochemical assays of blood are performed on Day 20. The state of the energy metabolism is assessed by the most significant intermediates such as ATP, lactate, pyruvate, and malate in protein-free extract prepared from the homogenates of heart, liver and brain. The results of the assay are compared to controls. When the content of the said substances is changed, the effect of ademethionine on the energy metabolism and prooxidant-antioxidant system in dichlorethane-induced hepatitis is evident.
