热休克蛋白70对人肺腺癌细胞顺铂耐药性的影响

摘要

Objective To investigate the expression of heat shock protein 70 (HSP70) induced by Geranylgeranylacetone(GGA)and synergistic effect on resistance to Cisplatin in SPCA-I cell line. Methods GGA-induced group and the control group were enrolled. The expression of HSP70 at protein and mRNA levels was analyzed respectively by immunofuorescence and reverse transcription polvmerase chain reaction (RT-PCR). Flow cvtometry was used to determine the effect of Cisplatin on cell apoptosis. Results Compared to the control group, HSP70 expression in both protein and mRNA levels in the GGA-treated group increased. The resistance rates to cisplatin in the control group was 9.70 ±0.63; in the 10, 50, 100,500, 1000 μmol/L GGA-induced groups it was(7.97 ±0. 14)%,(7.78 ± 0.69) %, (6.37 ± 0.15) %, (6.63 ± 0. 93) %, (4.22 ± 0.06) % (P < 0. 05). Flow cytometry assay showed that compared with the control group, GGA-treated cells dramatically decreased. Conclusions GGA affects on the expression of HSP70 in SPCA-I cell line. Up-regulation of HSP70 can signifcantly increase the sensitivity of Cisplatin.%目的 探讨用替普瑞酮诱导人肺腺癌细胞SPCA-1中热休克蛋白70(HSP70)的表达,并观察其对顺铂耐药的作用与联系.方法 1×106株人肺腺癌SPCA-1细胞完全随机分为5个不同剂量的替普瑞酮处理组和对照组,各5 ×105株,对照组加入磷酸盐缓冲溶液处理,替普瑞酮各组分别加入10、50、100、500、1000μmol/L的替普瑞酮处理.采用免疫荧光、逆转录聚合酶链反应方法检测各组细胞中HSP70mRNA和蛋白的表达;流式细胞仪法测定在顺铂作用下的细胞凋亡率,分析其在SPCA-1细胞对顺铂耐约的作用.结果 与对照组相比,细胞中HSP70、蛋白水平的表达均明显升高.流式细胞仪结果显示:在顺铂作用下,对照组细胞凋亡率为(9.70±0.63),替普瑞酮为10、50、100、500、1000 μmol/L时的细胞凋亡率分别为(7.97±0.14)%、(7.78±0.69)%、(6.37±0.15)%、(6.63±0.93)%、(4.22±0.06)%;替普瑞酮处理的各组细胞凋亡率均明显低于对照组(均P<0.05),且呈浓度依赖性.结论 替普瑞酮可上调SPCA-l细胞中HSP70表达,HSP70的高表达可增加SPCA-1细胞对顺铂的耐药性.
