“桂枝与白芍”药对化学成分UPLC-Q/TOF-MS分析

         

摘要

Objective To develop an ultra-performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometric (UPLC-Q/TOF-MS) method to analyze the chemical constituents in the drug pair of "Cinnamomum cassia Presl and Paeoniae Radix Alba".Methods Agilent 1290 UPLC machine and YMC-Triart C18 column (100 mm × 2.1 mm,1.9 μm) were adopted to separate ingredients;the mobile phase was acetonitrile (A)-0.1% acetic acid solution (B) with gradient elution;the column temperature was 30℃;the inject volume was 5 μL;the detection wavelength was set at 254 nm;the flow rate was 0.3 mL/min;the mass spectrometer AB 5600 with an electrospray ionization (ESI) interface was under positive or negative mode for data collection.Results 8 compounds were identified by reference substance,13 compounds were qualitative identified according to the MS/MS information,and the belonging of the main constituents had been confirmed.These compounds included six kinds of organic acid such as gallic acid and protocatechuic acid,tentotal paeoniflorin monoterpenes like paeoniflorin and albiflorin,and coumarins.Conclusion Determination of chemical composition of "Cinnamomum cassia Presl and Paeoniae Radix Alba" by UPLC-Q/TOF-MS in order to clarify the "Cinnamomum cassia Presl and Paeoniae Radix Alba" drug on the basis of the material to provide strong evidence.%目的 采用液质联用技术(UPLC-Q/TOF-MS)对“桂枝与白芍”药对中的化学成分进行定性分析.方法 采用Agilent 1290 UPLC和YMC-Triart C18(100 mm×2.1 mm,1.9 μm)色谱柱进行快速物质分离;流动相为乙腈-0.1%冰醋酸梯度洗脱;温度:30℃;进样量:5μL;检测波长:254 nm;流速:0.3 mL/min;AB 5600质谱使用ESI离子源分别在正离子与负离子模式下采集数据.结果 结合对照品确认了8个化合物,通过质谱信息与相关文献数据推测13个化学成分,对其进行成分归属.其中主要含有没食子酸、原儿茶酸等6种有机酸,芍药苷、芍药内酯苷等10种芍药总苷单萜类化合物及香豆素类成分.结论 采用UPLC-Q/TOF-MS确定“桂枝与白芍”药对中的化学成分,为阐明“桂枝与白芍”药对的药效物质基础提供有力的证据.
