醇沉4组分芦荟多糖通过不同给药途径对SD大鼠深Ⅱ度烫伤创面愈合过程中纤维连接蛋白含量的影响

         

摘要

目的 研究醇沉4组分芦荟多糖对SD大鼠深Ⅱ度烫伤创面修复过程中皮肤愈合率及皮肤组织中纤维连接蛋白(FN)含量的影响,探讨芦荟多糖促进烫伤创面愈合的可能作用机制.方法 大鼠背部用水浴烙铁法致伤,采用随机数字表法对大鼠分组.试验组用醇沉4组分(60%、70%、80%、90%比例)芦荟多糖水溶液采取外敷、灌胃及混合给药3种方式,对照组用生理盐水采取相同给药方式,2次/d.记录伤后3、7、14d创面愈合率,应用酶联免疫吸附试验(ELISA)测定创面皮肤组织中FN含量.结果 与对照组比较,试验组愈合率明显升高,80%、90%芦荟多糖组于烫伤后14d创面愈合率明显高于对照组(P<0.01).FN含量在第7天达峰值,之后逐渐下降.伤后3、7、14d试验组创面FN含量均高于对照组,80%、90%芦荟多糖组FN含量于第7、14天显著高于对照组(P<0.01),80%芦荟多糖组FN含量最高.给药方式上外敷优于灌胃,混合给药优于外敷给药.结论 80%芦荟多糖混合给药(外敷及灌胃)烫伤大鼠,促进伤口愈合的疗效最佳.芦荟多糖使烫伤创面皮肤组织中FN含量增加,促进创面皮肤愈合,为芦荟多糖制剂在烧烫伤方面的临床应用提供了实验依据.%Objective To study the effects of aloe polysaccharide in four components of alcohol precipitation on the healing rate of skin and the contents of fibronectin (FN) during healing process in SD rats with scalded wound surface of deep Ⅱ degree,and to investigate the possible mechanism of aloe polysaccharide in promoting the healing of scalded wound surface.Methods The back of rats was injured by iron in boiling water,and the rats were grouped by random digits table.Experimental group was applied with aloe polysaccharide aqueous solution in four components of alcohol precipitation (60%,70%,80% and 90%),with 3 administration routes of external application,gastrogavage and mixing administer;control group was applied with physiological saline by the same administration routes,twice a day.The healing rate of wound surface was recorded after scalding for 3,7,14 d,and the content of FN in wound surface was determined by enzyme linked immunosorbent assay (ELISA).Results Compared with control group,the healing rate in experimental group was increased significantly,and the healing rates in 80%,90% aloe polysaccharide groups after scalding for 14 d were significantly higher than that of control group (P < 0.01).The contents of FN reached peak in seventh day,then they were gradually decreased.The contents of FN in wound surface after scalding for 3,7,14 d in the experimental group were all higher than those of control group,the contents of FN in 80%,90% aloe polysaccharide groups after scalding for 7,14 d were significantly higher than that of control group (P <0.01),the content of FN in 80% aloe polysaccharide grou p was the highest.In way of administration,external application was better than gastrogavage,and mixing administer was better than external application.Conclusion The efficacy of 80% aloe polysaccharide by mixing administer (external application,gastrogavage) in scalded rats in promotingthe healing of wound is the best.Aloe polysaccharide makes the increase of FN in scalded wound surface,promote the healing of wound surface,and provide experimental basis for the clinical application of aloe polysaccharide preparations in burn and scald.
