复方甘草酸苷注射液对B16鼠黑素瘤细胞黑素生成的影响

         

摘要

Objective: To investigate the effect of compound glycyrrhizin injection on melanogenesis and mechanism of B16 murine melanoma cells. Methods:The influence of compound glycyrrhizin injection on the growth of B16 murine melanoma cells and the protein level and enzyme activity of tyrosinase, melanogenesis and melanosome transport was measured by CCK8, flow cytometry, NaOH splitting methods, dopa hydrocar-bonylation and western-blotting technique, respectively. Results:( 1) There was no influence on the growth of B16 murine melanoma cells when the concentration of compound glycyrrhizin injection was lower than 30μL/mL;The growth rate of B16 murine melanoma cells was retarded when the concentration was higher than 50 μL/mL;The cells were greatly inhibited when the concentration reached 500μL/mL. ( 2) The melanogen-sis and the protein level of tyrosinase were stimulated when the concentration of compound glycyrrhizin injec-tion was between 10 μL/mL and 200 μL/mL;The melanogenesis was greatly stimulated when the concentra-tion was 50 μL/mL, and the tyrosinase activity was increased in a concentration-dependent manner, which was peaked when the concentration was 100 μL/mL or 150 μL/mL. Conclusion: Compound glycyrrhizin in-jection can stimulate the melanogenesis and melanosome transport, and increase the tyrosinase activity.%目的::研究复方甘草酸苷注射液对B16鼠黑素瘤细胞黑素的影响及其机制。方法:通过CCK8、流式细胞术、NaOH裂解法、多巴氧化法和Western-blotting技术检测复方甘草酸苷对B16鼠黑素瘤细胞生长,黑素合成与转运及酪氨酸酶活性和蛋白含量的影响。结果:(1)复方甘草酸苷注射液浓度低于30μL/mL,对B16鼠黑素瘤细胞生长无明显影响;浓度大于50μL/mL,细胞生长速率减慢;浓度达到500μL/mL,细胞生长明显受抑制。(2)浓度在10μL/mL到200μL/mL,黑素的合成和酪氨酸酶的活性均增加。黑素的合成在复方甘草酸苷注射液浓度为50μL/mL时增加最明显;且酪氨酸酶活性呈浓度依赖性增加,在浓度为100μL/mL和150μL/mL时达到最高。结论:复方甘草酸苷注射液有促进黑素瘤细胞黑素的合成与转运及增加酪氨酸酶活性的作用。
