高效液相色谱法测定木糖醇产品中的木糖醇含量

摘要

用高效液相色谱面积外标法测定木糖醇产品中的木糖醇含量,以超纯水为流动相,采用Phynomenex公司的Rezex系列Ca2+型糖醇柱(带保护柱)做分离柱,选用示差折光检测器,控制分离温度为75℃,分离流速为0.6mL/min,对木糖醇产品进行测定,然后以峰面积对质量浓度做标准工作曲线,再由试样中组分的峰面积求出试样中组分的实际浓度,经试验表明,6次平行样的峰面积相对标准偏差为0.55%,5次加标回收率为99.64%,其相对标准偏差为0.87%,在质量浓度为0.000515g/100mL时具有很好的线性关系.此法简便快速,准确度高,适合于对大量样品的分析.%A method of determination of Xylitol concentration by HPLC area external standard is discussed.Distilled water was used as mobile phase,Rezex series of Ca2+sugars and Sugar Alcohols column made by Phynomenex company was used as separation column.Xylitol products with RI detector were determined at separation temperature 75℃ and separation flow rate 0.6mL/min.The standard work curve was made by peak area and mass concentration.the real concentration of Xylitol was calculated by the comparison with peak area in the standard curve.The results showed that relative standard deviation of the peak area in six parallel samples was 0.55%,the coefficient of recovery detected by five times was 99.64%,its relative standard deviation was 0.87%.When the concentration was 0.000515g/100mL,it had very good linear relation.The method was simple,rapid,accurate and fit for large quantitives sample analysis.
