In order to investigate process of extraction and purification, and hypoglycemic effects of Momordica charantia ( MC) stem, bitter stems were filtered by 60 mesh sieve and extracted twice at temperature 70℃ for 3h. The solvent extraction was 60% ethanol , solid-liquid ration was 1: 15. The crude extracts were then purified twice by AB-8 macro-porous resin . The extracts samples were analyzed by high performance liquid chromotography (HPLC) and the content of sapanin in extracts was tested by colorimetry. The extracts were used to treat hyperglycemic rats and the damaged in-sulinoma cell RIN-M5F in vitro. The effects of MC stem extracts on the metabolic parameters (serum glucose, insulin, glycogen, SOD activity and Malonedialdehyde (MDA) levels) of hyperglycemic rats and the effects on RIN -m5F cells were observed. The results showed that the saponin content of bitter stem was 83% after extraction and purification. Its polarity was similar to ginsenoside Rgl; there was a significant increase in serum insulin and glycogen, and a decrease in serum glucose and MDA in treated rats. The extracts also exhibited obvious protection and reparation on RIN-M5F cells.%研究苦瓜茎皂苷类物质的提取纯化工艺及降糖功效.在浸提温度为70℃,浸提时间3h,浸提溶剂为60%乙醇溶液,料液比1∶15的条件下对过60目筛的苦瓜茎粉浸提两次,粗提物经两次AB -8大孔吸附树脂纯化后得苦瓜茎皂苷,行HPLC分析并采用比色法检测提取物中皂苷的含量.采用苦瓜茎皂苷对高血糖小鼠及损伤的胰岛瘤细胞RIN - M5F细胞模型进行干预,观察高血糖小鼠血糖、胰岛素、肝糖原、SOD活性、MDA含量的变化,及损伤的RIN - M5F细胞的修复情况.结果表明,提取纯化后的苦瓜茎皂苷含量达到83%,与人参皂苷Rgl极性接近;经苦瓜茎皂苷灌胃后的高血糖小鼠血糖降低,血清胰岛素含量增高,肝糖原增加,丙二醛(MDA)含量降低;对损伤的RIN - M5F细胞有修复作用.
