双指标优化微波酶法辅助牛大力薯中多糖提取

         

摘要

The polysaccharides of Radix Millettiae speciosae contains a variety of physiological activity,which is the main functional components of its water extract.According to whether is there strong the root structure,Radix Millettiae speciosae can be divided into root and tuberous root.In this paper,the extraction process of polysaccharides was studied by microwave enzymatic method.The optimum extraction conditions that were determined by the yield of crude polysaccharides and the purity of polysaccharides were as follows:pectinase dosage 2.2%,enzymolysis temperature of 55 ℃,enzymolysis time of 2h,microwave time of 70s,the yield of crude polysaccharide was 1.24%,and the purity of polysaccharide was 50.88% by using glucose-sulfuric acid colorimetric method and glucose as control.It provides the data base for the utilization and the purification of polysaccharides about the tuberous root of Millettia speciosa Champ.%牛大力多糖具备多种生理活性,是牛大力水提物主要功能成分.牛大力又根据是否为块根状结构分为牛大力薯和牛大力根.采用微波酶法辅助对牛大力薯中多糖提取工艺进行了优化,以粗多糖得率和多糖纯度双指标确定了最佳提取条件为:果胶酶用量2.2%,酶解温度55℃,酶解时间2h,微波时间70s,制备粗多糖得率1.24%,采用苯酚一硫酸比色法、以葡萄糖作对照测定多糖纯度为50.88%.为牛大力薯中多糖的纯化及利用提供了数据基础.
