二氢杨梅素的制备及其对酪氨酸酶的抑制研究

         

摘要

The optimal technological conditions were determined by orthogonal experiments,and dihydromyricetin was separated from total flavones by the column chromatography-recrystallization. L-tyrosine and L-dopamine were chosen as substrate respectively, the inhibition of tyrosinase were investigated respectively. The results indicated that the optimal extraction conditions were as follows; volume fraction of ethanol was 60% , ratio of solid to solution was 1: 30 (g/mL) , extraction temperature was 40 ℃, time was 40 min, the ultimate yield of total flavones was 46. 54%. The yield and purity of dihydromyricetin were obtained by the column chromatography-recrystallization were 41. 12% and 99. 12% .respectively. The activities of monophenolase and diphenonase were obviously inhibited by dihydromyricetin, and the inhibition rates were 95. 87% and 69. 01% .respectively.%通过正交实验确定超声波提取藤茶中总黄酮的最佳工艺条件,用柱层析-重结晶法从藤茶中分离得到二氢杨梅素.选用L-酪氨酸、L-多巴为底物,分别测定二氢杨梅素对酪氨酸酶的单酚酶、二酚酶的抑制率.结果表明,提取的最佳工艺条件:提取溶剂是体积分数为60%的乙醇,料液比1:30(g/mL),提取温度40℃,提取时间40 min,总黄酮得率为46.54%.柱层析-重结晶法分离纯化得到的二氢杨梅素得率为41.12%,纯度为99.12%.二氢杨梅素对单酚酶、二酚酶均有显著的抑制作用,抑制率分别可达95.87%,69.01%.
