目的 制定一种以槐角为原料,适用于工业化生产制备染料木素的最佳工艺.方法 以槐角为原料,采取正交实验,确定最佳的槐角苷提取工艺;在单因素考察的基础上,采取正交实验,确定槐角苷水解得到染料木素的最佳条件.结果 槐角中槐角苷的最佳提取工艺是:80%乙醇、煎煮120 min、8倍量、提取3次;槐角苷最佳酸解工艺是:用2.5 mol?L-1盐酸乙醇液酸解,料液比为1:15,酸解温度为90℃,酸解时间为2.5 h.结论 该方法 经济适用,是可用于大批量生产的制备方法.%Objective To develop an optimum industrial preparation method for genistein from Fructus Sophorae. Method The optimal extracting condition of sophoricoside is to set up by orthogonal experiment from Fructus Sophorae, and on the basis of single factor experiment , the optimum reaction conditions of acid hydrolysis of sophoricoside were to establish by orthogonal experiment. Result By orthogonal experiment, the optimal extracting condition is add 8 times amount of 80% EtOH and to boil for 120 min and totally to extract for 3 times. The optimum reaction conditions of acid hydrolysis of sophoricoside were as follows:concentration of HCL 2. 5 mol · L-1 ,temperature 80 ℃ ,time 150 min,the ration of the original feed to HCL ethanol solutions 1:5. Conclusion This method is reasonable and feasible for industrial preparation of genistein.
