蛋白质二硫键异构酶参与血栓形成的研究进展

         

摘要

血栓形成可引起心、脑、肺、肾等重要脏器缺血,甚至危及生命。现代凝血理论已证实蛋白质二硫键异构酶在生理性、病理性凝血反应中发挥重要作用。蛋白质二硫键异构酶具有氧化还原、异构化及分子伴侣的作用,能够催化二硫键与巯基之间的互变反应,对参与血栓形成的多种蛋白具有调控作用。近年来关于蛋白质二硫键异构酶对血栓形成调节研究的不断深入,其抑制剂成为抗凝药物研发的新热点。现主要就其作用机制存在的争议展开综述。%Thrombosis can cause heart, brain, lung, kidney and other important viscera ischemia, even life-threatening.Modern co-agulation theory has confirmed that protein disulfide isomerase( PDI) in physiological and pathological blood coagulation reaction plays an im-portant role.PDI has three catalytic activities including, thiol-disulfide oxidoreductase, disulfide isomerase and redox-dependent chaperone, to proteins which participate in thrombosis have regulation effect.In recent years,the study of PDI of regulating thrombosis goes deeper, the inhibitors of it has become a new way of antithrombotic drugs.In the present review, we discuss its origin and its specific mechanism.
