颞叶癫痫发生不同时期的大鼠海马神经元GluR6和GABAAR-α1表达量的变化及其相互关系

     

摘要

目的 探讨颞叶癫痫大鼠海马神经元在不同时期GluR6和GABAAR-α1的表达量的变化,分析其与癫痫发生和发展的相互关系.方法 通过脑立体定向微量注射技术,向右侧大鼠海马CA3中心区注入KA以建立颞叶癫痫大鼠模型(KA组),采用同样方法向该区注射等量的生理盐水作为对照(对照组);视频监控大鼠癫痫发作的行为学特点;用Western blot方法检测海马神经元GluR6和GABAAR-α1表达量.结果 与对照组相比,KA组在癫痫发作急性期,海马神经元GluR6和 GABAAR-α1的表达均显著减少(n=5,P<0.05);在静止期,KA组海马神经元GluR6和 GABAAR-α1几乎没有表达(n=5,P<0.01);在慢性期,KA组海马神经元GluR6和 GABAAR-α1的表达均显著升高(n=5,P<0.05).结论 大鼠海马神经元GluR6和 GABAAR-α1表达量的异常可能是癫痫发生的原因之一.%Objective To investigate the variations of GluR6 and GABAAR-α1 subunits expression in different phases of temporal lobe epilepsy (TLE) in rat hippocampal neurons, and to analyze their relationship between the variations and the generation of epilepsy. Methods The TLE model rats ( KA group) were induced by kainate acidc(KA) microinjecting into the center of the CA3 region of the right hippocampus with stereotaxic technology.The control rats ( NS group) were injected with the same volume of saline solution in the same way as KA group.Video was used to monitor the behaviors of the rats and Western blot was used to detect the expression of GluR6 and GABAAR-α1 subunits in rat hippocampal neurons. Results Compared with the control group, the expression of GluR6 and GABAAR-α1 subunits in hippocampal neurons significanthy decreased in the acute phase of the KA group ( n = 5, P < 0. 05 ); There was almost no expression of GluR6 and GABAAR-α1 subunits in hippocampal neurons in the silent phase in the KA group (n =5, P <0. 01 ) ;The expression of GluR6 and GABAAR-α1 subunits in hippocampal neurons significantly increased in the chronic phase in the KA group ( n = 5, P < 0. 05). Conclusion The variations of GluR6 and GABAAR-α1 subunits expression may be involved in the generation of epilepsy.
