采用恢、保回交群体和集团分离分析(BSA),筛选了860个10mer随机引物,找到了与甘蓝型油菜波里马细胞质雄性不育系(POl cms)育性恢复基因(Rf)连锁的两个RAPD标记AHl9690和AI16830。它们位于Rf的一侧,与该基因的遗传图距分别为5.9cM和13.6cM,两标记间的遗传图距为7.8cM。这两个RAPD标记的发掘,为进一步利用分子标记辅助选育Pol cms新恢复系打下了基础。%Bulked segregant analysis was used to identify random amplified polymorphic DNA(RAPD) markers linked to the fertility restoring gene for the Polima cytoplasmic male sterility (Pol cms) in rapeseed (B. napus L.). Totally 860 random 10-mer primers were screened on the DNA samples of fertile and sterile bulks. Two primers, AH19 and AIl6, produced repeatable polymorphic bands between the paired bulks. DNA samples from individual plants of a back cross population (1141A/Hui5200)//1141B)were then used as DNA samples template for PCR amplification with these two primers.RAPD data was analyzed with program of Joinmap.The results indicated that the RAPD markers, AH19690 and AI16830 were linked to the Pol cms Rf gene at one side, with a map distance of 5.9 cM and 13.6 cM, respectively.The distance between the two markers was 7.8 cM.
