Complex peptide mixtures from enzymatic digests in proteomics,natural peptide libraries produced by microorganisms,complex synthetic combinatorial peptide or nonpeptide libraries can be screened efficiently using a sophisticated set-up for high-throughput HPLC-MS analysis established in our laboratories.In comparative studies using silica-based monolithic and packed capillaries hyphenated with high resolution FTICR-MS,the monoliths show a superior performance compared to packed beds of small-diameter beads [1,2].Silica-based capillary monoliths with bimodal pore size distribution possess a tailored adjustment of macro-and mesopores in the highly porous silica structure.As a result a combination of high separation efficiency and high loading capacity due to high surface area is realized which can not be provided by any type of particulate phase material.Moreover,coupling to the FTICR-MS allows the mass detection with highest sensitivity and resolution as well as MS"experiments for sequence analysis.
