Probiotic including lactic acid bacteria (LAB) are living microorganisms which are beneficial to human and animal health. In this study, LAB had been isolated from palm sap (Borassus flabellifer Linn.) origin Kupang, East Nusa Tenggara. LAB was identified based on 16S rDNA and applied for antibacterial analysis for selection of probiotic candidate. The result showed the number of LAB in palm sap range 3.2 × 10~8 cfu·mL~(-1) to 12.7 × 10~8 cfu·mL~(-1). Total of 36 strains were successfully isolated and identified. LAB was associated to palm sap were Leuconostoc mesenteroides (20 isolates), Lactobacillus plantarum (five isolates), Fructobacillus durionis (nine isolates), and Fructobacillus fructosus (two isolates). Antibacterial analysis on the isolates showed Leu. mesenteroides and L. plantarum have strong antibacterial activity (80 % to 100 %), inhibit six pathogenic bacteria (Escherichia coli, Pseudomonas aeruginosa, Listeria monocytogenes, Bacillus subtilis, Staphylococcus aureus, and Salmonella enteritidis) and F. durionis, F. fructosus have a weak antibacterial activity, inhibit about 11 % to 92 % to pathogenic bacteria. The isolates L. plantarum strain (Su-ls36, Su-ls45, Su-ls52, and Su-ls53) recommended for probiotic candidate. The isolates have strong antibacterial activity, survive in low acidity (pH 3), bile tolerance and ability to grow in high temperature (45 °C) also producing dietary enzymes such as amylase, protease, and phytase.
