Optimization on aqueous enzymatic extraction conditions of pine seed protein by response surface method

Hydrolysis process of aqueous enzymatic extracted pine seed protein was studied. Alcalase was selected as enzymolysis enzyme, the extraction rate of total protein was used as index. Single factor experiments and response surface experiment were applied to analyze the factors. Factor s contribution rate of were as follows: enzyme amount > hydrolysis temperature > the ratio of solid to liquid > pH > enzymolysis time. The results showed that the optimum conditions were: enzyme amount 1.9%, hydrolysis temperature 55, enzymolysis°C time 2.2h, the ratio of solid to liquid 1:5, pH8.8, total extraction rate of pine seed protein can reach 87.92%. Further more, transmission electron microscopy was applied to analyze the changes in the microstructure of pine seed with extend of enzymolysis time.
