In conclusion, multi-IMAC-HLB method is a competitive method to the widely used SCX-IMAC method. It could achieve more phosphopeptide identifications and give higher percentage of phosphopeptide detection, with much less starting materials. The sample preparation with IMAC-RP also requires less time and has a relatively straightforward procedure.
