1. Availability of multiple SIL analogs, deuterated vs. other SIL analogs, efficiency of extraction, purity of extracts, fold change of marker, and the anticipated need to dilute study samples are all things to consider when choosing between surrogate matrix and surrogate analyte approaches. 2. Quantitation of endogenous amino acids by surrogate matrix and surrogate analyte approaches agree well with quantitation by standard addition. 3. Acceptable accuracy and precision were achieved by both approaches. 4. Good dilutional linearity was shown for endogenous analyte with surrogate matrix. 5. Parallelism by surrogate matrix can require more extensive method development, while parallelism by surrogate analyte is accomplished by response balance. 6. Response balance is a time-consuming process.
