The present invention relates to a method for increasing the recovery of amino acid in serum or plasma and a quantitative analysis method for free amino acid in serum or plasma using a liquid chromatography-mass spectrometer (LC-MS/MS). According to the present invention, the recovery of amino acid which is water-soluble in a serum or a plasma sample can be increased through pretreatment for removing phospholipids that cause matrix effects interfering with the ionization of a subject to be analyzed by using liquid-liquid extraction methods before deproteinization. In addition, LC-MS/MS enables rapid and accurate analysis without separate derivatization reactions and the method can be useful as an aid for diagnosis and examination.
