Determination of microcystin microcystin-LR, Microcyatin-RR and Microcystin-YR in bovine rumen contents by LC/MS/MS

Microcystins are cyclic peptide toxins produced by cyanobacteria and are commonly found worldwide in freshwater and brackish water. Livestock mortalities have been linked to cyanobacteria toxins. A number of cattle deaths have been reported in the Prairies of Canada due to acute microcystin ingestion. The rapid and accurate analysis of microcystins in bovinerumencontentsis important for investigating the cause of cattle death and monitoringexposure of microcystinsfor risk assessment. Our laboratory developed a liquid chromatography tandem mass spectrometry (LC/MS/MS) method for the identification and quantification of micro cystin-LR, microcystin-RR and microcystin-YR in rumen contents after solid phase extraction using Oasis HLB cartridges. The method LOD for MC-LR and MC-RR was 0.05 μg/L and 0.1 μg/L for MC-YR. The limit of quantification was 0.1 μg/L for all the three analogous. The method is suitable for fast an d accurate identification and quantification of microcystins in bovine rumen contents.
