A General Approach for Glycoprotein Analysis: Glycopeptide Analysis by LC/MS and Glycan Permethylation Profiling

A glycoprotein characterization platform has been developed using 5 model glycoproteins and applied to analyze B/Malaysia/2506/2004 hemagglutinin including site occupancy and semi-quantitative analysis of glycoforms. The predicted glycosylation sites were found to be occupied in agreement with NetNGlyc when transmembrane is accounted for. CN3D alignment allows placement (but not 3D orientation) of glycans which should allow predictions of affects of glycosylation on epitopes (cell substrates). Automated informatics software is needed to assign glycopeptide identities accurately and reproducibly.
