Metabolic Profiling in Plasma samples of Patients Before and After Ketogenic Diet Therapy by Monolithic-C_(18) LC/(+)ESI-MS

Monolithic C_(18) chromatography was applied to underivatized human plasma samples successfully. Data processing/analysis software tools for the global metabolomic profiling of ten healthy female and male adults before and after a ketogenic diet therapy were evalated. As shown in the PCA plot, significant differences were found between females and male samples. Five percent of the total number of masses distinguished the diet for the female samples, and a similar number for the males. Carnitine and acylcarnitines profiles for all the samples under study were provided. Data base search, which was constrained by the two ppm mass tolerance-principle, was performed by the METLIN and the HMDB databases. Future work includes targeted metabolomics on putative biomarkers and further qualitative identification.
