In vivo and in vitro studies on the correlation between methylation and acetylation were undertaken. LC-MS experiments indicate that methylation of K4 and K36 in H3 has significant effects on global changes of acetylation levels in histone H3, but little effects on acetylation levels in H4, H2A, and H2B. LC-MS/MS experiments showed increased levels of K4 methylation correlated with an increase in the acetylation of K9 and K14 in H3, while the acetylation level of K56 in H3 decreased in the JHD2 DELTA strain. In contrast, K4 methylation showed little effect on K56 acetylation in RPH1 DELTA compared to WT.
