The natural vitamin E analogues (a-, (3-, y-, and d-tocopherols, a-, p-, y-, and 8-tocotrienols, a-tocopheryl phosphate) differently modulate signal transduction and gene expression. These events are the result of the modulation of specific enzymes involved in signal transduction, such as protein kinases C, protein kinase B, tyrosine kinases, 5-lipoxygenase, phospholipase A2, protein phosphatase 2A and diacylglycerol kinase, In HMC-1 mastocytoma cells, the tocopherols inhibit with different potency proliferation and induce apoptosis via inhibition of protein kinase B activity. In THP-1 monocytes, the tocopherols inhibit oxLDL-induced CD36 scavenger receptor expression via inhibition of the PBK/PKB/PPARy signaling pathway In this review it is suggested that the modulation of protein-membrane interactions and enzyme translocation to the plasma membrane may represent a common theme for vitamin E action.
