Fab' fragments derived from the ovarian carcinoma-specific monoclonal antibody OV-TL 16 were dimerized using a PEO (poly(ethylene oxide)) crosslinker. The in vitro binding and uptake of this (Fab')_2-PEO construct by OVCAR-3 human ovarian carcinoma cells was investigated and compared to that of unmodified OV-TL 16 antibody. It was determined that the binding constant of the construct is somewhat lower than that of unmodified antibody (7 x 10~7 M~(-1) vs. 2 x 10~8 M~(-1)). However, the in vitro rate of uptake of the construct by OVCAR-3 cells was enhanced relative to the native antibody.
