The present invention relates to a vector encoding a wild-type or truncated form of the erythropoietin receptor (EpoR) to promote T cell survival and proliferation. In particular, the invention is an anti-CD19-41ΒΒ-003ζ chimeric antigen receptor (CAR) and cleavage type, demonstrating significantly higher anti-leukemic activity in vivo with increased increase in ex vivo over CAR-T cells. An example is a bicistronic vector expressing EpoR. The invention also describes methods for producing cells expressing the vector and the use of these cells to kill CD19 + tumor cells.
