The present invention provides a method for evaluating the quality of a dephosphorylation reagent containing alkaline phosphatase, and a method for detecting a target nucleic acid using a dephosphorylation reagent evaluated to be of high quality by the evaluation method. In order to achieve such an object, the present invention is a method for evaluating the quality of a dephosphorylating reagent, and the following steps: (1-1) A step of preparing a dephosphorylation reagent containing alkaline phosphatase and a peptide fragment derived from the alkaline phosphatase; and (1-2) the content of the peptide fragment relative to the content of the alkaline phosphatase. Provided is the method including a step of evaluating that the quality of the dephosphorylation reagent is high when the ratio is equal to or less than a predetermined reference value.
